n-3 fatty acids and rosiglitazone exerts additive effects on muscle glycogen synthesis in mice fed a high-fat diet

Combined effects of n-3 polyunsaturated fatty acids (n-3 PUFA) and rosiglitazone were studied on a murine model of obesity and insulin resistance. Male C57BL/6 mice were fed for 5-20 weeks (i) high-fat (HF) diet (35 g fat/100 g diet); (ii) HF diet with n-3 PUFA concentrate replacing 15% dietary lipids; (iii) HF diet with 10 mg rosiglitazone /kg diet; (iv) combination of both n-3 PUFA and rosiglitazone. The combination exerted additive effects in prevention of obesity and associated low-grade adipose tissue inflammation, dyslipidaemia, and insulin resistance, while inducing adiponectin, suppressing hepatic lipogenesis and decreasing muscle ceramide concentration. The improvement in glucose tolerance reflected a synergistic stimulatory effect of the combination treatment on muscle sensitivity to insulin. The combination treatment may reduce dose requirements and hence the incidence of adverse side-effects of rosiglitazone therapy.
