Chemical derivatives of docosahexaenoic acid (DHA) in the prevention and reversal of obesity in mice fed a high-fat diet
The efficacy of α-substituted DHA derivatives as lipid-lowering, antiobesity, and antidiabetic agents was analyzed. C57BL/6 mice were given a high-fat (35% weight/weight) diet or high-fat diet with 1.5% of lipids replaced with α-methyl DHA ethyl ester, α-ethyl DHA ethyl ester (Substance 2), α,α-di-methyl DHA ethyl ester, or α-thioethyl DHA ethyl ester for 4 months. Substance 2 prevented weight gain, and independently of its mild effect on food intake it also prevented development of pathological consequences of obesity, namely glucose intolerance, fasting hyperglycemia, elevated plasma levels of insulin and lipid metabolites, lipid accumulation in non-adipose tissues as well as adipose tissue inflammation. Substance 2 also induced weight loss and improved glucose intolerance in dietary obese mice. Therefore, this DHA derivative has a potential to be used for the treatment of obesity and associated metabolic disorders.
