Abstract |
New model nucleosides bearing another nucleobase pointing out to the major groove of DNA will be designed and synthesized as analogues of transition states of DNA methyltransferases and DNA glycosylases. These nucleosides will be then phosphorylated to trophosphates and transformed to protected phosphoramidites that will be enzymatically and chemically incorporated to oligonucleotides and DNA for studying of mechanism and inhibition of the target enzymes. This interdisciplinary project will embrace the interface between organic chemistry, biochemistry and chemical biology and will heavily utilize methodologies of all these fields. |
