Core Facilities


Genomics and Bioinformatics Facility

The facility was established in late 2005 after purchase of the Affymetrix GeneChip System and was initially operated by staff from the Department of Mouse Molecular Genetics . Since January 2007 it became an independent unit which provides full chip microarray services (expression analysis, genotyping and resequencing), real-time quantitative PCR service and high-throughput methods using the liquid-handling robotic equipment. The services are provided not only to the research groups at the Institute of Molecular Genetics but also to other academic institutions in the Czech Republic as well as abroad. The core facility is equipped with two microarray platforms: Affymetrix GeneChip System and Illumina BeadStation 500, real-time PCR cycler Roche LC480 and JANUS robots from PerkinElmer, and also with instruments for assessment of quality and quantity of processed samples (spectrophotometer Nanodrop and capillary electrophoresis Agilent Bioanalyzer 2100).

Monoclonal Antibodies Facility and Cryobank

The new cryobank (capacity over 350 000 samples) was established in January 2007 and is used for storage of cell lines, mouse sperm, and embryos in liquid nitrogen. The storage containers are supplied with liquid nitrogen from a tank {capacity of 6000 litres}. It is equipped with an independent power supply in case of emergency. The samples can be stored in liquid nitrogen or nitrogen vapour. The operation, diagnostics and maintenance of storage containers is fully automated and controlled. The operating parameters of storage containers and safety of the whole unit are checked by a monitoring system with GSM and web interface outputs. The monoclonal antibody service laboratory performs mycoplasma cell culture testing and provides complete service for preparation of new monoclonal antibodies, including: immunization of mice, fusion of mouse spleen cells with myeloma tumour cells, primary antibody production screening (using ELISA test), cloning by limiting dilution or agar clonation, secondary antibody production screening (using e.g. ELISA), production of monoclonal antibodies into the cell culture supernatants, generation of a hybridoma cell bank.

Flow Cytometry and Light Microscopy Facility

The facility provides methodological and instrumental background for flow cytometric and fluorescence microscopy techniques. Core facility is opened for users from institutes of Academy of Sciences of the Czech Republic and for users from other institutions as well.

At present the facility is equipped with two analyzers - BD FACSCalibur and BD LSRII cytometers. The LSRII instrument has been upgraded with the yellow 561nm laser and is now a four laser (405nm, 488nm, 561nm and 633nm) instrument with 14 fluorescence detectors. This upgrade together with a large set of dichroic mirrors and bandpath filters made this instrument very flexible and capable to cover most of the flow cytometry applications. Both instruments have HTS (High Throughput Sampler) loader for high throughput analysis of samples directly from 96- or 384-well plates. The facility is also equipped with AutoMACS Pro (Miltenyi Biotec) magnetic separator for automatic rapid sorting of cells, as well as cell culture equipment.

Facility has been recently equipped with state of art microscopy instrumentation: inverted fluorescent microscope Leica DMI6000 with superfast scanner (Leica TCS SP5 AOBS TANDEM) and 8 excitation laser lines (405, 458, 476, 488, 496, 514, 561 and 633nm); Leica DMI6000 inverted microscope with TIRF illumination and 4 excitation laser lines (405, 488, 561 and 633nm); wide field inverted fluorescent microscope DeltaVision (Applied Precision) with 405 and 488 nm lasers for photo manipulation. All three microscopes have the climatic chambers allowing performing the experiments at 37°C in CO2 athmosphere. This instruments allows facility users to use wide range of microscopy techniques including FRET, FRAP, time lapse experiments, membrane studies, vesicle transport, etc.

Media preparation and glass washing unit

The service unit offers preparation of tissue culture media and solutions, preparation of bacteriology media and plates, sterilization of solutions and material (vapour sterilization, filtration), distribution of FBS, transfection agents, glass and plastic washing, decontamination of GMO waste, organization of working cloth washing.

Animal Facility

An array of breeding types is available at the animal facility: conventional breeding, breeding in isolated cages for animals in quarantine from accredited suppliers, special quarantine for unverified animals from non-accredited suppliers, special SPF breeding with individually ventilated breeding cages. The special quarantine enables import of animals from the entire world and their subsequent transfer to SPF conditions. Besides various mouse strains the animal facility breeds also laboratory chicken, duck, hamster and fish.

X-ray radiation facility

The facility is equipped with an orthovoltage x-ray instrument T-200 (Wolf-Medizintechnik) for regulated radiation of cells and mice.

Information Technologies

The IT department ensures the administration of computer network in the domain img.cas.cz, administration of institutional servers (dns, mail, web) and storages including data backup. The IT department provides installation and registration of computers and printers to the computer network, hardware purchases and consultancy, support for users of PC and MAC platforms. The IT department also operates the audio-visual equipment in the conference hall. Furthermore, the IT department negotiates and maintains multilicensing contracts for software.
