Topic Bio- and Air-tolerant C-C Bond Formations via Organometallic Ruthenium Catalysis

Abstract

In spite of the Diaspora of organometallic reactions across the field of preparative organic synthesis in the last 30 years, the use of organometallics towards the formation of C-C bonds under physiological conditions in presence of biomolecules has been widely neglected. We recently formulated an attractive and unexplored hypothesis, based on the notion that organometallic reactions compatible with aqueous aerobic environments might be viewed as a direct entry into a variety of chemical species (metal-containing as well as metal free) with potential bioactivity. Searching through such species generated in situ, i.e. under physiological conditions in immediate presence of target biomolecules, will allow for direct investigation of binding or reactive events otherwise difficult or even impossible to realize. This non-traditional potential of organometallic reactions suggests the most exciting window of opportunity and calls to be validated. The goal of this project is to design relatively simple experimental situations that will serve as proof-of-principle examples.
