Abstract

Novel types of modified nucleoside triphosphates will be prepared by direct cross-coupling reactions in aqueous solutions and will be incorporated to DNA by polymerases. A combination of enzymatic ligations and cleavages by endonucleases will be used in the constructions of target sequences of single stranded oligonucleotides and DNA duplexes for bioanalytical applications (sensors of DNA hybridization, minisequencing etc.) and applications in chemical biology (studying of binding of transcription factors etc.). This interdisciplinary project will embrace the interface between organic chemistry, biochemistry and chemical biology and will heavily utilize methodologies of all these fields.
