Topic Synthesis of fluorescently-labelled nucleoside triphosphates and enzymatic construction of labelled nucleic acids for the study of their interactions with proteins

Abstract

Novel modified nucleoside triphosphates will be prepared and their polymerase catalyzed incorporations to oligonucleotides and DNA will be studied. The base-modifications will focus on environment-selective fluorescent groups (solvatochromic fluorophores, molecular rotors). Interactions of these modified DNAs with selected DNA-binding proteins (e.g. restriction endonucleases, DNA methyl-transferases, transcription factors etc.) will be studied.
