Novel redox or fluorescent DNA labeling for bioanalytical applications
Nucleobases bearing an electrochemical or luminiscent marker in a suitable position could be used in bioanalysis. Novel types of nucleotides containing such nucleobases bearing redox active molecules (complexes of transition metals or organic functional groups) or fluorescent labels (substituted biaryls etc.) linked via conjugated linkers are designed and prepared in our group. Polymerase incorporation of the corresponding modified nucleoside triphosphates is used for construction of oligonucleotide probes that serve as sensors of hybridization - collaboration with Prof. M. Fojta (Institute of Biophysics, AS CR, Brno). Also in collaboration with the Fojta group, bioanalytical techniques (i.e. minisequencing) based on incorporation of different redox labels for electrochemical detection are developed, as well as a "multicolor" (multipotential) redox coding of DNA.
