Topic Development of small molecules targeting c-myc oncogene

Abstract

Binding of small molecules to promoter G-quadruplexes has been shown to modulate transcription of the associated genes. This project focuses on c-myc oncogene, overexpression of which is linked with wide range of human cancers. The work will include chemical synthesis, purification, and characterization of novel type of heterocyclic G-quadruplex ligands able to bind and to stabilize c-myc G-quadruplex DNA. Part of the work will be accomplished in close collaboration with an interdisciplinary team conducting research of biological effects of the newly synthesized ligands including assessment of their ability to bind to G-quadruplex and inhibit expression of c-myc oncogene on both mRNA and protein levels. The research will be conducted at the Institute of Organic Chemistry and Biochemistry, CAS.
