Topic Synthesis of modified nucleoside triphosphates and enzymatic construction of functionalized nucleic acids for the study of their interactions with proteins

Abstract

Novel modified nucleoside triphosphates will be prepared and their polymerase catalyzed incorporations to oligonucleotides and DNA will be studied. The base-modifications will focus on molecular probes (redox-active or fluorescent groups) and chemically reactive groups for bioconjugation (i.e. cross-linking with proteins). Interactions of these modified DNAs with selected DNA-binding proteins (e.g. restriction endonucleases, DNA methyl-transferases etc.) will be studied.
