Topic Construction of functionalized nucleic acids bearing selected switchable or triggerable reactive groups

Abstract

Polymerase synthesis of base-modified oligonucleotides and nucleic acids (DNA and RNA) from base-functionalized nucleoside triphosphates (NTPs) will be studied. The starting modified NTPs will be prepared by aqueous cross-coupling reactions. The base-modifications will focus on chemically or photochemically switchable and/or triggerable reactive groups. Further post-synthetic chemical modifications of the functionalized nucleic acids (including bioconjugations) and chemical or photochemical switching or triggering of the reactivity and/or cross-likning with proteins will be also studied.
