Topic Identification of compounds disrupting protein-protein interaction and cap binding in influenza A polymerase

Abstract

The student will focus on a high-throughput method for screening of small compounds disrupting the protein-protein interaction between two subunits of influenza RNA polymerase, and blocking cap-binding by PB2 polymerase subunit which appears to be  promising drug targets. The student will further investigate the 3D structure of complexes between protein and active compounds by X-ray crystallography for characterization of their binding mode. He/she will acquire knowledge on biophysical methods such as AlphaScreen, surface plasmon resonance (SPR), X-ray crystallography, microcalorimetry or differential scanning fluorimetry (DSF).
