Topic Chemical biology tools for imaging, targeting and specific killing of tumor cells
Faculty PřF / LF1 UK / VŠCHT
Abstract

We have recently developed a modular system of polymer “synthetic antibodies” (iBodies) that allow for visualisation and  isolation of target molecules based on specific interaction between the protein target and small molecule ligand (Šácha et al., 2016).  iBodies: Modular Synthetic Antibody Mimetics Based on Hydrophilic Polymers Decorated with Functional Moieties. Angew. Chem. Int. Ed. Engl. 55, 2356-2360).  We continue to apply this tool for a variety of targets including membrane and cellular proteases, carbonic anhydrases and immune cell receptors. The successful applicant will develop novel iBodies against medicinally relevant targets, will design or select specific low-molecular ligands of these targets and analyze the interaction of corresponding macromolecule conjugates with the target. The final aim of the project is to design and test iBodies capable of recruitment of cytotoxic immune cells to tumor cells.

We look for highly motivated candidate enjoying work in multidisciplinary team capable of working at the interface between molecular biology, biochemistry, structural biology and medicinal chemistry. The methods actively used involve recombinant DNA cloning, protein expression and characterization, X-ray and NMR structural biology, immunochemistry and transgenic mice.
