The preparation of new protected nucleoside phosphonic acids and their phosphonothioate congeners as monomers for solid phase oligonucleotide synthesis. The elaboration of novel so far unavailable methodology allowing the combination of phosphoramidite and phosphotriester methods at the preparation of both phosphorothioate and phosphonate/phosphonothioate internucleotide linkages, and its verification at the synthesis on the solid phase.
