Project title: Chemical biology for drugging undruggable targets

Brief description: The objective of a research agenda conceived in this way is the development of new potential therapeutics of cancers, viral diseases and metabolic disorders on the principle of specific inhibition of so-called "undruggable" interactions of biomacromolecules, on which classical medicinal chemistry cannot reach. The research will be focused on three key types of biomolecular interactions: protein-protein; protein-membrane and protein-DNA or protein-RNA. With the use of a multidisciplinary approach combining the structurally- and molecularly biological characterization of the target biomacromolecules and their interactions, bioinformatics and molecular modelling, design and chemical synthesis of a series of potential inhibitors (including libraries of sequences of modified peptides and oligonucleotides), detailed testing of the inhibition of targeted biomolecular interactions, or in vitro selection methods of their identification from the libraries of sequences, and study of the cytostatic and antiviral activities, or physiological effect of the tested substances, substances with the highest activity and lowest toxicity will be selected for further preclinical studies.
Within the project, research teams will be particularly enriched with newly acquired local and foreign talented doctoral candidates, post-docs and junior researchers. Project will ensure the development of the existing infrastructure mainly in the area of theoretical calculations and modelling, experimental studies of protein interactions, automatic testing of substances, microscopy, sequencing RNA, DNA and in the area of chromatographic separation. Project activities will support the international research cooperation with world recognized research institutions: Catholic University in Leuven, Johns Hopkins University and the pharmaceutical company Gilead Sciences, Inc. - Realisation schedule: 1. 1. 2018 - 31. 12. 2022

Project title: PharmaBrain

- Brief description: The project is aimed at the research of the glutamatergic neurotransmitter system that controls wide range of brain processes. This system also takes part in normal brain development, while the dysfunction of this system plays the key role in many neuropsychiatric diseases. The project is focused on its role in clinical practice as well as in experimental models. The research in this area has a long tradition in all participating institutions and also a high reputation among the professionals worldwide.

Project title: Carbon allotropes with rationalized nanointerfaces and nanolinks for environmental and biomedical applications
- Acronyme: CARAT

- Brief description: The main motivation for the proposed project CARAT is a radical change in the modus operandi of interdisciplinary research in highly actual research area focused on carbon nanomaterials. The project formulates completely new strategy of the material design, with the ultimate aim of rationalizing communication channels (interfaces and nanolinks) to the environment in accordance with the nature of the chemical bond of the respective carbon allotrope. At the same time the project targets the establishment of an expert team with a deep engagement in all relevant sectors of the academic and application interest. Based on previous successful cooperation at national and international level, the team members are able to speak the same language, so maximize the profit from the shared expertise and know-how. Internationally, an important outcome of the project is the establishment of the Joint Laboratory of Detonation Synthesis, where groundbreaking experiments will be conducted in the worldwide unique facility by team of experts in all fields linked. In this context, it is expected that the Czech Republic will establish itself as a world-power in the preparation of detonation nanodiamond with precisely defined parameters and hybrid materials based on graphenoids and/or nanodiamond for biomedicine, energy storage and environmental applications.
