Modified nucleoside triphosphates with unnatural nucleobases for enzymatic synthesis of modified DNA
Abstract
The project will deal with the study and development of the chemical synthesis of diverse modified nucleoside triphosphates derived from d5SICS and dMMO2 nucleosides, which are used in the extension of the genetic alphabet. New derivatives bearing useful functional groups will be designed and synthesized aiming at the modification of DNA in the major groove. Substrate activity of these dNTPs with diverse DNA polymerases will be studied and they will be used for site-specific modification of DNA or in enzymatic synthesis of hypermodified DNA.
