Surface plasmon resonance biosensor for analysis of complex biological samples

Year: 2013

Prof. Jiří Homola, Ph.D., DSc.

We have developed a surface plasmon resonance (SPR) biosensor employing a new referencing method that allows suppressing the effect of non-specific adsorption, which is the main limitation for the application of label-free biosensors in the analysis of bodily fluids. We have demonstrated that the developed SPR biosensor enables detection of the carcinoembryonic antigen (cancer biomarker) with higher accuracy and lower biological variability with respect to conventional detection approaches.

New referencing method: principle and experimental data

(Left) Principle of the new referencing method. (Right) Error of the reference-compensated sensor response in human plasma for three different plasma samples (squares, circles and triangles, respectively), two different concentrations of CEA - 200 ng/mL (upper plot, open symbols) and 500 ng/mL (lower plot, solid symbols). The new method and conventional referencing method using four different reference receptors (antibodies) are presented on the left (in yellow) and right columns, respectively.
