Genetic Toxicology and Epigenetics

Research Department

Group photo of the Department of Genetic Toxicology and Epigenetics team

The department is focused on the mechanisms of toxic effects of engineered nanoparticles as well as combustion originated fine and ultrafine particles. Major attention is paid to the interaction of particles and their components with biomolecules in human cell lines. Currently, DGTE is focused on the role of ultrafine particles in the etiology of neurodegenerative diseases. The methods include various tests of cytotoxicity, the detection of reactive oxygen species and oxidative damage of DNA, proteins and lipids (ELISA), analyzes of genotoxicity (Comet assay, DNA adducts) and clastogenic effects (micronucleus test and cytogenetic analysis by FISH), as well as analyzes of protein expression (western blotting) and DNA repair activity (NER and NHEJ). To get more insight into the mechanism of biological effects of particles or their extracts, the toxicogenomic methods (microarrays and next generation sequencing) and RT-PCR are used. Most of these tests are also available to external users from research, universities, industry and academia via the Research Infrastructure NanoEnviCz (www.nanoenvicz.cz).

Important results

Maternal Diet Quality and the Health Status of Newborns

The study evaluated the quality of the maternal diet and the health status of newborns. The results showed insufficient intake of vegetables, dairy products and fiber. The intake of the most persistent organic pollutants did not exceed limits, only the intake of perfluorinated substances exceeded the limit for 7.8% of women. Birth weight had a positive relationship with protein intake. Neonatal oxidative stress had a positive relationship with DDT intake and a negative relationship with protein intake.

Graphical abstract of the study and its results. First, the nutritional quality of the maternal diet was evaluated from the detailed food records (1) and chemical analysis of food samples contamination by persistent organic pollutants was performed (2). Subsequently, the relationships of these two factors with birth weight (3) and neonatal oxidative stress (4) were studied.
