1 Cell surface and intra-acrosomal sperm proteins.
1.1 To study of the role of selected cell-surface proteins and intra-acrosomal proteins of spermatozoa in binding sperm to the glycoprotein network of zona pellucida of oocytes.
1.2 To determine the difference in localization and/or exposition of sperm proteins, cytoskeletal and heat shock proteins in healthy and subfertile men.
RESEARCH PLAN
2 Study of molecular mechanism of reproduction in mammals.
2.1 Characterization of sperm surface proteins and proteins secreted by the male reproductive tract and study of their function in individual steps of the mammalian reproduction process.
2.2 The origin of seminal plasma proteins in the male reproductive organs.
3 Tyrosine phosphorylation, cytoskeleton.
To examine the unique regulation of protein tyrosine phosphorylation during capacitation and after acrosome reaction together with localization of these proteins in mammalian spermatozoa, as well as their connection with the cytoskeleton.
4 Effect of selected pollutants on cell and mammalian reproduction.
To determine impact of chosen pollutants on cells in vitro (toxicity, viability and function) as well as on mammalian organisms in vivo (reproductive organs, gametes and reproduction). The expression of selected gene connected with normal and pathological state will be used as biomarker for reproductive organ damage. For detection of gene activity the RT-PCR method will be used.
5 A detailed study of spermatozoa in fish model.
To determine motility, ATP consumption, acrosome enzyme activity and integrity in two groups of model species of Chondrostean and Teleostean fish.
6 Practical application.
To develop methods and tools (monoclonal antibodies) for objective and sensitive diagnosis of male infertility and to uncover the toxicological effect of selected drugs on fertility.
